HEK293 cells derived from human embryonic kidney cells are widely used for producing various therapeutic proteins, virus-like particles (VLP) in vaccines, viral vectors in cell and gene therapy, or for related scientific research. During the production and purification of these products, impurities of host cell protein (HCP) will inevitably be introduced. These impurities can reduce the efficacy of the product and cause adverse toxicity or immune reactions. Therefore, it is necessary to reduce HCP impurities to the lowest practical level.

This kit uses the experimental principle of double-antibody sandwich enzyme-linked immunoassay (ELISA) to detect HEK293 HCP residues. The ELISA method is simple, highly sensitive and objective. To detect HEK293 HCP residues, add the HEK293 HCP standard (36713-B) and the sample to be tested to the ELISA plate pre-coated with anti-HEK293 HCP antibody (36713-A). Then add diluted biotin-labeled HEK293 HCP detection antibody (36713-C), and finally add Streptavidin-HRP (SA-HRP) (36713-D) to form antibody+antigen+antibody‐Biotin+SA ‐HRP complex. After washing the plate, add TMB chromogenic solution (36713-H) to develop color. TMB is transformed from colorless to blue under the catalysis of HRP enzyme and finally transformed into yellow under the action of stop solution (36713-I). The shade of yellow is positively correlated with the amount of HEK293 HCP detected in the sample. This kit can be used for optimizing the purification process of biological products, controlling impurities in the intermediate process, and detecting the release of final products
